Literature Search of Protein Ligand Binding
and Enzyme Kinetics

Find a recent article from a scientific journal (the ACS Journals are available
online from on-campus) about measuring the interaction between a protein and a
ligand. Make a copy of the article to annotate and hand in with your
answers. For each question, indicate where in the article you found your
answer.
Note the following about the article:
- The equation used to analyze the data.
- Whether bound protein or bound ligand was measured.
- Whether cooperative interactions were encountered.
- The specificity of interaction between protein and ligand.
- Whether the binding interaction is considered to be high-affinity.
Also find an article measuring constants associated with enzyme kinetics.
Identify about the experiments:
- what kinetic model was used to analyze the data.
- if the experiment measured product formed or reactant consumed.
- if the pH dependence of the reaction was measured, or how the pH of
the reaction was selected.
- the significance of the measured kinetic constants.
- whether covalent modifications of the enzyme are necessary for the
measured activity.
- if the enzyme is allosterically controlled.
These articles should be studied in lieu of class attendance. The articles
with associated answers will be due in class on Wednesday, September
26.
